Genetic characterization of species with restricted distributions, limited historical records, or uncertain population status represents a major challenge for biodiversity research and conservation, particularly when accessible natural populations for direct sampling do not exist. In this context, the present study proposes molecular characterization of the mitochondrial cytochrome c oxidase subunit I (COI) gene in Puerto Rican coquí species using specimens preserved in scientific collections, applying DNA barcoding methodologies adapted for historical material. The study is based on the analysis of fixed and preserved specimens, primarily in ethanol and formalin, with associated provenance data including locality, collection date, and previously documented morphological characteristics. DNA extraction will follow standardized DNA barcoding protocols incorporating specimen-specific adjustments for degraded tissues, including prolonged digestions and optimized amplification conditions.
